To improve the efficiency of mRNA production workflows, cap analogs have been developed that allow for the addition of a 5’ cap during mRNA synthesis. These co-transcriptional capping methods can produce abundant mRNA bearing a 5’ cap in a single reaction step.
While co-transcriptional capping IVTs are more streamlined than their traditional counterparts, wild-type T7 RNA Polymerase does not efficiently incorporate many of these cap analogs leading to capping efficiencies far below those required for therapeutic use.
The Codex® HiCap RNA Polymerase is an engineered T7 RNA Polymerase designed to address the challenges of efficiently producing therapeutic-grade mRNA at scale. Compared to wild-type T7, Codex® HiCap offers a higher affinity for cap analogs leading to equivalent or better yields and higher capping efficiencies.
