Considerations when using kits for plasmid DNA production
- For unstable plasmids (e.g. AAVs), use recombinant-deficient host strains for plasmid propagation
- Be gentle when performing alkaline lysis steps to reduce genomic DNA contamination
- Avoid exceeding 25 % of the volume of the growth vessel to ensure maximum aeration
- Don’t store biomass in -20 C freezers that undergo thawing cycles
- When working with large plasmids (> 20 kb), consider increasing the volumes of the suspension, lysis and neutralization buffers
